  • Bio-inspired Enzyme Mimics Built from the Self-Assembly of De Novo Designed Short Peptide
        After millions of years of evolution, existing enzymes are composed of at least hundreds of amino acid residues. Polypeptides with long sequences are thought to be able to form complicated structures upon molecular folding and thus possess recognition, accumulation, and catalytic functions. However, this consensus may be challenged by catalytic short peptide assemblies.
        Based on the structural and functional characteristics of natural enzymes, we have designed and constructed several kinds of enzyme mimics, including carboxylate hydrolase, peroxidase, laccase and phosphate hydrolase, through the self-assembly of de novo designed short peptide. We systematically summarized the basic approaches and strategies for the design and construction of peptide-based enzyme mimics, including: (1) introducing the catalytic amino acids of natural enzymes into short peptide sequence, (2) reconstructing of the electron transfer system of natural enzyme, and (3) replicating the key structure of natural enzyme. In addition, we focused on the effects of the binding of catalytic cofactors (metal ions or organic molecules), the assembly degree of peptide and the supramolecular structure on the catalytic functions of these enzyme mimics.

        This work provides new ideas for the design and construction of enzyme mimics inspired by natural enzymes, it also expands the understanding of the role of short peptide assemblies in the evolution of “primitive enzymes".
